PG - 197-204 LID - S0145-305X(14)00204-3 [pii] LID - 10.1016/j.dci.2014.07.023 [doi] AB - In the present study, we found that recombinant grass carp IL-1beta (rgcIL-1beta) simultaneously up-regulated grass carp IL-1beta (gcIL-1beta) and TGF-beta1 (gcTGF-beta1) expression via NF-kappaB and MAPK signaling in grass carp head kidney leukocytes (HKLs), promoting us to clarify whether TGF-beta1 is an effective antagonist in IL-1beta expression and activity. Our results showed that a stimulation of gcIL-1beta on its own expression was noted within 6 h, but gcTGF-beta1 neutralizing antibody prolonged gcIL-1beta autostimulation up to 12 h, indicating a possible inhibitory role of gcTGF-beta1 in regulating gcIL-1beta effect. This notion was reinforced by the fact that recombinant grass carp TGF-beta1 (rgcTGF-beta1) could impede rgcIL-1beta-induced gcIL-1beta gene expression and secretion in a reciprocal manner. Further studies revealed that rgcTGF-beta1 was able to attenuate rgcIL-1beta-induced mRNA expression of its own receptor signaling molecules and the activation of NF-kappaB. By contrast, rgcIL-1beta significantly amplified rgcTGF-beta1-mediated gcTGF-beta1 type I receptor (ALK5) expression and Smad2 phosphorylation in the same cell model. Taken together, these data shed light on an intrinsic mechanism for controlling inflammatory response by the reciprocal interaction between TGF-beta1 and IL-1beta in teleost.
