PG - 4762-4767 LID - 15609 [pii] LID - 10.26355/eurrev_201808_15609 [doi] AB - OBJECTIVE: Tumor necrosis factor-alpha (TNF-alpha) participates in the regulation of the whole process of pregnancy. Matrix metalloproteinases-2 and -9 (MMP-2 and MMP-9) play important roles in the process of trophoblast invading decidua. This research aims to determine the role of IFN-gamma in TNF-alpha, MMP-2, and MMP-9 of abortion rats. MATERIALS AND METHODS: The rats were divided into control, abortion model, gestation, and IFN-gamma group. Abortion rats model in IFN-gamma group were treated by IFN-gamma at low and high doses upon abortion model. Serum and tissue TNF-alpha, MMP-2, and MMP-9 expressions were detected by ELISA and immunohistochemistry. RESULTS: The level of TNF-alpha was significantly elevated, while MMP-2 and MMP-9 were statistically declined in the serum and decidual tissue of rat from model group (p < 0.05). Of note, the expression of TNF-alpha was further increased, whereas MMP-2 and MMP-9 reduced in IFN-gamma high dose group (p < 0.05). CONCLUSIONS: We demonstrated that in abortion rats, TNF-alpha was overexpressed, while MMP-2 and MMP-9 were reduced declined in the serum and decidual tissue. The treatment with a high dose of IFN-gamma further upregulated TNF-alpha expression and decreased MMP-2 and MMP-9 levels, aggravating the severity of rat abortion.
