However, its role in laryngeal squamous cell carcinoma (LSCC) remains unknown. AIM: The study aimed to explore the effect of MIAT/miR-147a/BCOR axis on LSCC progression. METHODS: The expression pattern of MIAT, miR-147a and BCOR in LSCC samples and cells was identified through qRT-PCR. The proliferation of LSCC cells was assessed by colony formation assay and CCK-8 assays. Transwell assays were implemented to test the migratory and invasive abilities of LSCC cells. Proteins associated with migration and epithelial-mesenchymal transition were probed in transfected LSCC cells by western blot. The interaction of miR-147a with MIAT or BCOR was analyzed by luciferase reporter assays, RNA pulls down assays and Ago2-RIP assays. RESULTS: High MIAT expression was closely correlated with unfavorable prognosis. MIAT knockdown inhibited cell proliferation, migration, invasion and EMT progress in LSCC. MIAT acted as a miR-147a sponge to increase the expression of BCOR. Silencing of MIAT suppressed LSCC progression through miR-147a/BCOR axis. CONCLUSION: MIAT acts as an oncogene by controlling miR-147a/BCOR axis in LSCC. CI - Copyright (c) 2020 IMSS. Published by Elsevier Inc. All rights reserved.
